Luminesensor repressible recA408 promoter
recA promoter in 408 form.
recA promoter belongs to the SOS regulon family. In E.coli it promotes the transcription of RecA protein, which forms filament when bound to ssDNA and trigger the self-cleavage of LexA protein. LexA protein can also bind to recA promoter, inhibiting the transcription of the genes downstream, too.
However, with mutations in specific sites which LexA protein will bind to, recA 408 promoter will not be bound by LexA protein, but is compatible to LexA 408 protein with corresponding mutations in DNA binding site. So a bio-orthogonal promoter is formed because the influence of endogenous LexA protein is excluded.
