Design Notes
The par locus was deleted through excision of BBa_K524000 by SspI and StuI. The resulting product generated 3 fragments with blunt ends. One of the fragments contained the par locus and some junk sequences. The three fragments were resolved with gel electrophoresis and only the two larger fragments with the par locus were recovered. Upon ligation, some plasmids had resulted in this new BioBrick in the orignal pSB1C3 vector.
