Design Notes

The yeGFP part (BBa_K319039) from last year didn't have a stop codon so this tagged promoter should have a terminator sequence cloned downstream before using.

Source

The distal region of the Adh1 promoter was PCR amplified from a plasmid source of BBa_K319005 and using overlap extension PCR was fused to the amplified proximal region of the Gal1 promoter. The Gal1 promoter is a commonly used promoter in our lab and we amplified it from a plasmid source. yeGFP was PCR amplified from a plasmid source of BBa_K319039.
