Applications of BBa_S01010

We use this part in the device 2 in our team OUC-China in 2011( please see http://2011.igem.org/Team:OUC-China/Result/fv), However, when we ligate it with K084005, B0015, R0079. We see that the DNA length is much longer than we expected. The electrophoretogram is as below. The S01010 can act as a cinI AHLs gnerator with reporter ECFP, It can paly an important role in our device 2 in our team OUC-China in 2011,saving us a lot of time, as shown in http://2011.igem.org/Team:OUC-China/Result/fv,

The K084005, B0015, R0079 and S01010 (18-3-11-16) are supposed to add up to 3000, however the electrophoretogram result above shows that they surpass 3000 greatly.

then we sequence the S01010, find that this part is of quality problems indeed. It proves that S01010 is 3094bp (longer than its expected length 1631bp)and its actual sequence is as below,

Through sequence analysis, we can see that the S01010 has lost its necessary part C0076 and only includes RBS, E0020 and B0015 in the end. Its former sequence has been wrong, this may affect its E0020's expression, so no ECFP is expressed in device II when detected under fluorescence microscope.
