Applications of BBa_K533009

This part is constructed as a tool to instruct E. coli chemotaxis toward a gradient of aspartate. Since it is originally one of the chemotaxis receptors of E. coli, the effect of chemotaxis cannot be seen until all the other chemoreceptors have been knocked out.

Dr. Mark Goulian's lab has characterized the chemotaxis property of this expression cassette in E. coli strain UU1250 (Δtsr-7028 Δ(tar-tap)5201 Δtrg-100 Δaer-1).

pPD10, the empty vector demonstrated no significant chemotaxis while both pPD12 and pMG65 exhibited chemotaxis towards aspartate gradient. In pMG65, the tar receptor is expressed from a constitutive strong trc promoter while in pPD12 tar is expressed from a very weak promoter inherent in lacI coding sequence. The results indicates that weaker expression of the chemoreceptor makes more sensitive chemotaxis (Derr, Boder et al. 2006).
