T7 RNA polymerase with tyrosine amber codons

T7 RNA polymerase with two tyrosine codons mutated to an amber (stop) codon.

Usage and Biology

The T7 RNA polymerase have two stop codons at the begening of its sequence. The protein cannot be expressed unless a tRNA amber supressor is expressed in the cell. (for instance K606034 for B. subtilis). This strategy can be use to create an AND gate. If an amplified signal is required, K606018 or K606035 can be used for reporting.
