T7 RNA polymerase with tyrosine amber codons

T7 RNA polymerases where two tyrosines were mutated to an amber (stop) codons.

Usage and Biology

The T7 RNA polymerases have two stops codons at the begening of its sequence. The protein cannot be expressed unless a tRNA amber supressor is expressed in the cell. (for instance K606034 for B. subtilis). This strategy can be use to create an and gate. If you want then the signal to be amplify, you can unse both K606018 or K606035 for reporting.
