T3 promoter drived amplifier+Agn43
This part can generate antigen 43 that can cause auto-aggregation under the regulation of T3 RNA polymerase. The T3 polymerase is also a specific activator of a series of T3 promoter that can regulate downstream genes.We designed this polymerase to be a system that is parallel to T7 polymerase driven expression. The amplifier is constructed by engineering PhiR73 and Po promoter together. PhiR73 is a phage derived activator that can activate Po promoter. We put antigen 43 downstream of Po promoter so that it will be driven indirectly by the expression of PhiR73. We then put PhiR73 under the regulation of T3 promoter, so that antigen 43 gene is now controlled by T3 promoter. In this way we have engineered a inductive expression part.
