pTet-rbs-chez_histag-double terminator
This part is an improved version of BBa_K094102. BBa_K094102 was designed to express CheZ protein, which is responsible for the dephosphorylation of CheY, a major effector in bacterial flagella rotation. cheZ gene is driven by the pTet promoter, whose expression is induced by adding anhydrotetracycline (aTc). The backbone of this biobrick is BBa_J61003. A strong double terminators lie between XbaI and SpeI sites.
Since lacking an antibody to recognize the expressed CheZ, we fused a His6-tag to the cheZ encoding region. Thus, the expression of cheZ can be easily detected by immunostainning and western blotting analysis. And this part has been demonstrated work as expected in our system (you can get more information in our wiki).
