SacB promoter - SpoVG RBS - MazE/F - Fd Terminator

SacB promoter is a sucrose induced promoter from Bacillus subtilis. MazEF is one of the most common Toxin-antitoxin systems (TA system), belonging to the kid family of the Class II TA classification system, and was first identified in E. coli. While mazF is a stable toxic protein capable of non-specifically cutting intracellular RNA and causing cell death, MazE is an unstable antitoxin that antagonizes MazF and can be degraded by the ATP-dependent ClpPa serine protease in cells. mazE is less stable than mazF, so cells must be able to survive when mazE is consistently expressed. And we needed a terminator to help us stop the above transcription, so we chose the fd terminator, which is the central terminator of bacteriophage fd. Additionally, we repeated the fd terminator as a strong terminator.
