Design Notes

"1. Different spacers derived from the article of synthetic promoters are placed between Inv AraI1 and AraO2 to ensure the original 193 bp interval present in natural E.coli is conserved. AraO2 is one bp upstream from TSS to explore different oppotunities. 2.Inversion of Ara operon with two AraI half sites reverses the original position of AraO. The inversion reduces the potential repressive effect due to dimeric AraC binding to AraI half sites in the Ara operon. 3.T-rich spacers results in nucleosome-free regions to reduce the potential impacts from neighbouring DNA.
