HMG-CoA synthase promoter (sterol regulated)
This part is a natural HMG CoA synthase promoter responsive to SREBP and when placed upstream of a gene makes it regulatable by the elements to which this promoter is sensitive.
Usage and Biology
3-hydroxy-3-methylglutaryl-CoA synthase (HMG CoA synthase) is an enzyme catalyzing the condensation of Acetyl-CoA and acetoacetyl-CoA to form 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA). There are two distinct HMG CoA synthases, the cytosolic and the mitochondrial form, encoded by two different genes. The reaction catalyzed by the cytosolic enzyme is a part of the biosynthesis of cholesterol. In mitochondria the same reaction is responsible for keton body formation. Sterol regulatory elements in the promoter region of the gene are responsible for transcriptional regulation. The natural HMG CoA synthase promoter was induced by Lipoprotein Deficient Serum (more details: see Cell Culture Notebook) in HeLa cells. Promoter activtiy was then roughly characterized analogous to REU by TECAN (automated plate fluorescence reader). For more natural promoters look at Part:BBa_K203105 and Part:BBa_K203107.
