Design Notes

BamHI is added before the ccdB sequence while HindIII is added after the ccdB sequence to facilitate separate vector concatenation and enzymatic identification recovery afterward, but there are no these two enzymatic sites in the whole sequence. The uploaded BBa_K4256166 sequence contains a lactose manipulator-related sequence to facilitate IPTG induction during experiments, but the full sequence for practical applications does not contain a lac operon or can be enzymatically excised away.
