Pspac promoter

This promoter can be induced by IPTG and it is tightly regulated by LacI. Wild type lac operator is replaced with a perfect palindromic "oid" operator sequence which increases the repression by LacI more than 2.5 fold and provide a To repress the promoter LacI needs to be expressed. Addition of IPTG will derepress LacI activity.

This biobrick is an enhancement of Biobrick BBa_K090501 which has the sequence from pmutin2. BBa_K143015 and BBa_I746665 biobricks have also the wild type lac operator.
