Design Notes
This toehold switch was designed according to the B-series toehold structure (Pardeee et. al., 2016). The A-series structure was designed to reduce translational leakage compared to older toehold switches (Green et. al., 2014). The binding site of the toehold switch was designed to bind to a conserved sequence in the BYDV genome, allowing it to sense the presence of this trigger sequence. This toehold has a downstream 21-nt linker sequence, which ends in the codon AAG. To prevent frame-shifting, the last G is omitted from the documented sequence and the junction site should have the sequence GATG instead of the standard AATG junction site.
