This part encodes a single guide RNA targeting one of the lsr2 paralogs ( lsr2A ) encoding the main xenogeneic silencer of Streptomyces . It has been designed to target different strains such as S. ambofaciens ATCC 23877, S. bottropensis ATCC 25435, S. venezuelae ATCC 10712 and S. rimosus ATCC 10970.
Xenogeneic silencers of Lsr2 family have been shown to silence the expression of specialized metabolite biosynthetic gene clusters (Gerke et al., eLife, 2019 - doi:10.7554/eLife.47691 ; Zhang et al., Genetics and Molecular Biology, 2021 - https://journals.asm.org/doi/10.1128/mBio.01077-21 ; Deng et al., RSC Adv, 2017 - DOI: 10.1039/c7ra06482a). Thus decreasing their expression has been associated with increased production of specialized metabolites including antibiotics.
