Spectinomycine selective cassette
This cassette allows the selection of Cyanobacteria transformant upon addition of spectinomycin into the media of culture. The sequence of the gene carrying the resistance is codon optimized for an expression into Synechcoccus elongatus.
Introduction
The SpecR cassette is composed by the promoter PaadA and the resistance gene specR (aadA). The resistance gene is based on the part (BBa_BBa_K1323012), but it was codon optimized for expression into S.elongatus.
This gene codes for a Aminoglycoside adenylyltransferase , which inactivate the spectinomycin by adding a adenyl group on it. Therefore, this cassette aload the selection of Yeast transformant upon addition of spectinomycin into the media of culture.
Part characterization
The part (BBa_K3930032) was used to select the right genomic integration of the part (BBa_K3930026). Transformants were plated onto BG11 + 10 μg.ml-1 spectinomycin Petri dish. The Toulouse INSA UPS team manage to select a transformant with this selection marker (for more details, check the (BBa_K3930026) part page)
