Design Notes
The main consideration is about how to obtain the functionally correct proteins from E.coli BL21,so we employed codon optimization to the sequence of the fusion protein. Another issue to consider is the linkage sequence between mamC and ZZ, which is refer to a recent research working on constructing a similar product as ours.
Source
The sequence of mamC comes from the genome sequence of Magnetospirillum gryphiswaldense MSR-1 and ZZ is the immunoglobulin-binding domain of staphylococcal protein A from the cell wall of Staphylococcus aureus. Both sequence were codon optimized to maximinze the expression in E.coli.For mamC (GeneBank AVM72836.1),you can visit https://www.ncbi.nlm.nih.gov/protein/AVM72836.1,and for ZZ(GeneBank:M74186.1),you can visit https://www.ncbi.nlm.nih.gov/nuccore/M74186.1 for more detailed information.
