MS2-responsive ribozyme switch (On to Off)

This part is a protein-responsive ribozyme switch that works in mammalian cell. It is designed to locate on the 3' end of an mRNA, after the coding sequence. Once mRNA is transcribed, the switch sequence on the mRNA will naturally form a secondary structure. This structure is designed as a combination of a ribozyme actuator and a MS2 aptamer sensor domain. When the MS2 coat protein is absent in the cell, the function of the ribozyme is not active so the secondary structure will not undergo self-cleavage. Therefore, the mRNA can be translated normally. Otherwise, once MS2 coat protein binds with the MS2 sensor domain, the ribozyme will be activated. Then the secondary structure will automatically cleavage and the polyA tail will be removed from the mRNA. By accelerating the degradation of mRNA, this part can regulate the expression of upstream gene(s).
