Anti-THC antibody fragment (ScFv), bound to EGFP at the C-terminal
Part encodes for an antibody fragment designed to detect delta-9-tetrahydrocannabinol (THC). The antibody fragment is linked to EGFP via a short GSGSGS linker on the C-terminal. The transcript is codon optimized for expression in E. coli. Additionally, an OmpA sequence proceeds the transcript, which serves as a per-plasmic localization signal. To function properly, the protein needs to form disulfide bonds, which it can only do in the oxidizing environment of the peri-plasmic environment. However, for best expression, we recommend Rosetta-gami2 for expression, which contains an oxidizing cytoplasmic environment.
