PsspB-gerAa

This part expresses in B.subsilis to increase the germinability of the spore.

Usage and Biology

The gerA operon from Bacillus subtilis encodes the spore’s germinant receptor for L-alanine. Overexpression of the tricistronic gerA operon will increase markedly the rates of germination of Bacillus subtilis spores with L-alanine,in particular at low L-alanine concentrations. The promoter for the B.subtilis sspB gene (PsspB) is more active than the endogenous gerAa promoter. It is also active during germination. This device consists mainly of the sspB promoter followed by the first 303bp of the gerA operon (first 303bp of the gerAa gene). Since B.subtilis exhibits accurate and efficient homologous recombination, a single cross-over event between the gerAa region in the BioBrick and the endogenous gerA sequence inserts the sspB promoter upstream of the spoVA operon,which promote the germination of Bacillus subtilis.
