TUDelft-Leiden 2014 |width='60%' valign='top'| We could not detect the coding sequence of mtrCAB by restriction analysis of part BBa_K1172401 or by sequencing the part BBa_K1172403 (mtrCAB with medium Promoter and medium RBS), which should contain the mtrCAB coding region of BBa_K1172401 according to Bielefeld 2013 iGEM team. TUDelft-Leiden 2014 iGEM team has created part BBa_K1316012 which contains the proper mtrCAB coding sequence on it and, therefore represents an improvement of this part.

Click here to see the picture of the gel. The first lane is smart ladder, sizes: 10000, 8000, 6000, 5000, 4000, 3000, 2500, 2000, 1500, 1000, 800, 600, 400 and 200 bp. The second lane corresponds to 5ul of BioBrick K1172401 digested with EcoRI and XbaI. Therefore, the expected size of the band on lane 2 is 7.4 kb (BioBrick K1172401 + pSB1C3 Backbone). The actual size is, however, about 4.2 kb. The rest of the bands are not relevant.
