Design Notes
The cloning of this part included adding a restriction site at the beginning of the mutant T7 RNAP: XbaI at the 5' end, which was added by PCR (this was done to suit the RNAP for the standards). There wasn’t any necessity to add the suffix restriction sites at the 3' because it was already at the end of the RNAP.
