pir gene

ori-gamma is part of the replication origin of the R6K origin. Initiation of replication at ori-gamma requires the pi protein in trans, which is encoded by the pir gene. Yet doubling the concentration of pi protein would effectively shut down the replication as well. Expression of pi protein is auto-regulated at its promoter region. Our construct design was to insert a CAP-binding site between the auto-regulating promoter and the CDS, by doing so we can halt replication and induce plasmid loss in nutrient (glucose) rich medium.
