DNA program

This part contains a DNA program - a specifically designed nucleotide sequence containing 6 zinc finger binding sites. The program was designed in such a way, that chimeric enzymes (fused with zinc finger domains) for synthesis of violacein bind to the sequence in the order required to produce the final product. The part contains binding sites for zinc finger proteins in the following order: Jazz - Blues - Zif268 - PBSII - HivC - Gli1.

We have bound five chimeric enzymes, encoded by Part:BBa_K323135 and Part:BBa_K323132 to the program sequence and have shown increased synthesis of violacein, in addition to a decrease in formation of side products, namely deoxychromoviridans.
