  • Without LuxI (or synthetic OHHL), RFP levels increase.

Experiment : checking BBa_I731012

  1. Picked and cultured the following stab and glycerol stocks in 2mL of LB:
    1. LB-Amp, BBa_I731012, (stab, received from iGEM)
    2. LB-Amp+0.2%Glu, BBa_K084007(plac+rbs+LasI(no LVA)), (JW1998)
    3. LB-Amp+0.2%Glu, BBa_K084012(plac+rbs+LuxI(no LVA)), (JW1998)
  2. Pre-Cultured at 37°C for 12h.
  3. Added 6.25% each of the pre-cultures to new LB medium.
    1. LB-Amp, BBa_T9002, BBa_K084012(plac+rbs+LuxI(no LVA)), BBa_K084007(plac+rbs+LasI(no LVA))
  4. Cultured at 37°C for 12h
  5. Mixed the sender cells BBa_K08400 and 7BBa_K084012 both with BBa_I1731012 at a 1:1 ratio.
    1. BBa_I1731012 alone.
    2. BBa_K084007:BBa_BBa_I1731012=1:1
    3. BBa_K084012:BBa_I1731012=1:1
  6. Culture 12h and observe results(follow picture).
