pGAL1

pGAL1 is a strong inducible promoter of GAL gene family, functioned as a transcriptional factor that is regulated by the level of galactose and repressed by the presence of glucose.

The galactose inductibility of this part has been widely used in heterologous protein expression. In our project, this part is utilized to construct a plasmid with Bax gene to fulfill the kill switch design with a logic gate induced by galactose in S.cerevisiae.


Usage and Biology

pGAL1 is an inducible promoter of GAL gene family, where GAL gene functions as a transcriptional factor that is regulated by the level of galactose and repressed by the presence of glucose.

Activator Gal4p and repressor Gal80p conditionally bind to the target DNA sequences to regulate the gene expression. In the presence of galactose, the removal of repressor activates Gal4p to contribute to a high level of transcription, whereas in the presence of glucose, Gal80p will bind to the activator that repress the gene transcription level.
