Design Notes

The design of RNF2 pegRNA includes the U6 promoter, spacer, guide RNA scaffold, rttpbs and polyA tail.

The original RNF2 pegRNA design containing spacer and rttpbs sequence was reported in Prime Editor 2 paper cited in References. With the help of and TWIST Bioscience we synthesized those and other necessary sequences, which were further pieced together with 65777 backbone in our lab.
