Design Notes

The design of RNF2 pegRNA includes the U6 promoter, spacer, guide RNA scaffold, rttpbs and polyA tail.

Source

The original RNF2 pegRNA sequence containing spacer and rttpbs was reported in Prime Editor 2 paper cited in References. We used those sequences and with the help of and TWIST Bioscience synthesized the parts, which were further pieced together with 65777 backbone in our lab.
