pBTK1001 part backbone
This is an entry vector from the Bee Microbiome Toolkit that is designed for initial part plasmid creation from PCR products or gBlocks. This plasmid and the PCR product are digested with BsmBI to form the initial part plasmid. Once they're in the pBTK1001 backbone, our parts are designed to be further digested with BsaI and integrated into an assembly vector such as pBTK1028 (BBa_K4579033). The first four bases and last four bases of the sequence for this entry vector are the overhangs generated after digestion with BsmBI. The entry vector plasmid contains a GFP dropout to allow for screening. This backbone was created by Peng Geng of the Barrick lab at the University of Texas at Austin.
