Taxadiene synthase is the enzyme which makes the precursor to paclitaxel, taxadiene, from GGPP. It is naturally occurring, found in the yew tree (genus Taxus). We specify that this taxadiene synthase is cytosolic rather than from the MEP pathway in the chloroplast1, which would require us to add export tags and make our engineering goal more difficult to reach. Taxadiene Synthase II2, a variation of taxadiene synthase from the Chinese yew has been found to have the highest output of paclitaxel out of any plant model, and is more effective in paclitaxel biosynthesis then the more well-known, Pacific Yew-derived taxadiene synthase3.
