EforRed validation system
BBa_K4256188 is a system used in experiments to verify the effect of the oxygen-controlled promoter HIP-1. The results of high expression of chromogranin under anaerobic conditions and low expression under normoxic conditions will verify the role of HIP-1. BBa_K4256188 combines Composite parts BBa_K4256155 and BBa_K4256177 and replaces the antitoxin protein sequence in BBa_K4256177 with the colored protein eforRed sequence. Under hypoxic conditions, the [4Fe-4S] cluster in the FNR protein is stabilized, prompting the FNR protein to form a homodimer, which can specifically bind DNA sites and initiate the expression of the colored protein eforRed downstream of the target promoter HIP-1, which can be observed in red; under normoxic conditions, the [4Fe-4S] cluster is disassembled and the FNR dimer is depolymerized. The initiation of eforRed transcription was inhibited and no red color could be observed. By this design, the effect of the oxygen-controlled promoter HIP-1 can be conveniently observed and verified.
