Design Notes
The polyhedrin promoter is a hyper-expression promoter that drives the expression of the polyhedrin gene found in baculoviruses. Due to its high expression power, the polyhedrin promoter may overwhelm a cell's protein production machinery when expressing multiple proteins, and deplete cellular resources along with a decrease in protein yield. Modifying the polyhedrin promoter to decrease its expression capacity can be used to optimize multiple protein expression yield ratios.

This promoter is part of the vector pFastBac used for Bac-to-Bac expression system. We obtained the plasmid pFastBac as a generous donation from the Airola lab at Stony Brook University.
