The coding gene of ACE2-PmrB recombinant protein.
We use plasmid pET28a to express this protein to test our SARS-CoV-2 detection device.
The polypeptide used in this project to mimic S protein is the 333 to 526 residues of S1 subunit, which is from the monomer of homologous trimer spike glycoprotein and is responsible for binding to ACE2 protein. The peptide only retained the binding function with ACE2 receptor, which is the RBD region of the combining point of ACE2 and S1 subunit of S protein. S1 protein subunit only retains the RBD region, which is the recognition region, but it takes both S1 and S2 protein subunits' participation for S protein to recognize ACE2, thus realizes plasma membrane fusion. Therefore, the residues we used has almost no risk to human body. We will not use SARS-Cov-2 and any samples that may contain it.
