The Level 1 plasmid “L1b” uses an ampicillin resistance for bacteria selection and its multiple cloning sites “RFP” is flanked by the Golden Gate restriction enzymes BsaI (GGAG/CGCT) and BpiI (GCAA/ACTA). In comparison to the original pICH47732 backbone, the “L1b” plasmid has a decreased size of more than 2000 bases.
