Uric acid-induced Uricase generator
This device could produce uricase only when there is uric acid. In the device, HucR is a repressor than can bind to HucO operator to inhibit the gene expression down stream HucO. In the presence of uric acid, the structure of HucR is altered and is no longer able to bind HucO, leading to the down stream gene expression. We inserted a uricase gene (URI) downstram HucO. In this case, uricase is expressed only when there is significant level of uric acid. To better visualize the gene expression, we include RFP(E1010) downstream HucR and a GFP(E0040) downstream URI. In our project, we express it in E. coli. It is hoped that the uricase-expressing E.coli can lower the uric acid concentration in human alimentary canal, so as to prevent gout disease.
