Tac Promoter (including two operator genes(lacO))


This promoter is a hybrid of two operons: the trp and lac operons. This promoter is inducible by IPTG and commonly used in Corynebacterium glutamicum for overproduction of proteins. Corynebacterium glutamicum constitutively produces the LacI protein, a strong Ptac promoter repressor which can bind with lac operator. In our experiment, we add two lac operators in order to regulate the expression of the racE<i/> gene to produce γ-PGA with different D/L-glutamate monomer ratios.
