Heat-shock-indued chromoprotein expression plasmid (pHeat)
This plasmid senses warm temperatures using the heat-shock protein 70 promoter (hsp70) and produces red pigment (meffRed) when activated. The warmer the temperature, the more red pigment that is produced. meffRed is tagged with a 6X-His tag, which facilitates Western blot analysis of meffRed production.
Dynamic range experimentation concluded that no meffRed is visibly produced after heat-shock at a variety of warm temperatures after 60 minutes; Western blot analysis also confirmed these results. Future experimentation could look at reverse transcription analysis to see if mRNA is being produced after heat-shock. If not, utilizing a composite part with a different heat-shock promoter is recommended.
