pGadA-MazF kill switch

Kill switch designed to reduce viability of E. coli upon contact with low pH environments in the range of normal rain drops (5-6).

pGadA promoter was characterized by team Dundee 2016 (BBa_K1962013) with GFP protein in different pH ranges. The GPF coding sequence was substituted by MazF, a endoribonuclease well characterized in kill switch approaches (1). This could limit replication of chassis for paratransgenesis in the environment, while allowing it to thrive inside the mosquito midgut.
