PsiR from Agrobacterium tumefaciens
This part is an E. coli codon optimized version of the PsiR found in Agrobacterium tumefaciens str. C58 (Atu4743).
Usage and Biology
PsiR is a predicted LacI family transcription factor with high affinity for D-Psicose. This means that PsiR will prevent transcription of the regulated promoters in the absence of D-Psicose by binding the consensus sequences before the Transcription Start Site.
Our team used this transcription factor together with the Agrobacterium tumefaciens PsiA or PsiR promoters (BBa_K2448010, BBa_K2448011) as well as with the pPsiTac1 synthetic promoter (BBa_K2448016) to build three Psicose biosensors (BBa_K2448025, BBa_K2448026 and BBa_K2448027). The results shows that PsiR behaved as predicted under and without induction.
