Constitutive E2 killing protein production
This design of this cascade system is similar to our other part BBa_K2447016, with the addition of constitutive expression of E2 Colicin (an DNAse). E2 killing protein activity is balanced off by the controlled expression of E2 immunity protein (IM2) via appropriate phosphate and temperature inputs.
This construct is useful to serve as a highly specific kill-switch where under normal circumstances (high phosphate concentrations or when the temperature is higher than 36 degrees), there would be constant production of E2 immunity protein to sequester the killing protein E2. Hence the cell would still survive. For other scenarios such as when the phosphate concentration is low and when temperature is lower than 36 degree, the kill-switch would be 'activated' where downstream production of E2 immunity protein will be down-regulated and causing over accumulation of E2 Colicin (since it is constitutive) and causing subsequent cell death.
