anti-STAR

Anti-STAR is the reverse complement of STAR (BBa_K1893013). It is identical to part of the STAR target sequence (BBa_K1893031), but much shorter in length and binds to STAR with greater affinity.


Usage and biology

The anti-STAR DNA sequence is identical to part of the 5’ end of the STAR target DNA sequence, but shorter in length. Anti-STAR was designed as an inhibitory RNA sequence for STAR. When transcribed, it produces a RNA product that is fully complementary to the transcribed antisense-STAR product. Anti-STAR has a higher affinity for STAR than the STAR target sequence, which allows it to sequester STAR and prevents it from binding to the STAR target sequence to activate transcription of a downstream coding sequence.


Anti-STAR was designed by using RNAstructure, a software tool that allowed us to predict a complementary RNA sequence to STAR that binds with higher affinity to STAR than it does to the full STAR target sequence.
