Applications of BBa_K2042004

BBa_K2042004 is a IPTG inducible promoter used for engineering of Pseudomonas putida. It belongs originally to the plasmid pSEVA224, which accomplishes with the pSEVA standards for cloning vectors.

iGEM Évry 2016 aimed to use it for the expression of Pct and PhaC genes, used for PLA biosynthesis. However, they did not succeed on cloning the operon and experiment with it in the bioplastic production context.

BBa_K2042004 was characterized, in consequence, by inserting GFP gene and analysing fluorescence data. Results have demonstrated that it is a leaky promoter. It was expected like that as its mechanism is through repression. With no or low levels of IPTG we have some gene expression. However, high concentrations can help increasing the expression, helping bioproduction optimization of compounds.
