Lac operator
A regulator sensitive to the Lac Repressor (encoded by the regulator gene LacI) and the catabolite activator protein (CAP).
The regulator contains two binding sites. The first is bound by the CAP protein, which is generally present in E. coli and is asocciated with cell health and availbaility of glucose. CAP contains a helix-turn-helix motif and binds major-groove DNA in the presence of the allosterif effector cyclic-AMP at the site, 5'-AATGTGAGTTAGCTCACTCATT-3'. The second site, 5'-AATTGTGAGCGGATAACAA-3', is bound by the LacI repressor protein. When the ispopropyl beta-D-1-thiogalactopyranoside (IPTG), which is an analog to allolactose, binds, LacI is inhibited.
In the absence of LacIl protein and CAP protein, this part promotes transcription. In the presence of LacI protein and CAP protein, this part inhibits transcription. LacI can be inhibited by IPTG. LacI is coded by BBa_C0010.
