TUDelft-Leiden 2014 |width='60%' valign='top'| We could not detect the coding sequence of mtrCAB by restriction analysis of part BBa_K1172401 or by sequencing the part BBa_K1172403 (mtrCAB with medium Promoter and medium RBS), which should contain the mtrCAB coding region of BBa_K1172401 according to Bielefeld 2013 iGEM team. TUDelft-Leiden 2014 iGEM team has created part BBa_K1316012 which contains the proper mtrCAB coding sequence on it and, therefore represents an improvement of this part.

  • Restriction Analysis of BBa_K1172401: Click here to see the picture of the gel. The first lane is smart ladder, sizes: 10000, 8000, 6000, 5000, 4000, 3000, 2500, 2000, 1500, 1000, 800, 600, 400 and 200 bp. The second lane corresponds to 5ul of BioBrick K1172401 digested with EcoRI and XbaI. Therefore, the expected size of the band on lane 2 is 7.4 kb (BioBrick K1172401 + pSB1C3 Backbone). The actual size is, however, about 4.2 kb. The rest of the bands are not relevant.
