Unbound DnaA fraction regulatory module

Upstream transcriptional activity leads to increased unbound fraction of DNA binding proteins.(Ref. Transcription through the roadblocks: the role of RNA polymerase cooperation. Epshtein et.al.EMBO J 2003). We have used upstream promoters pLac and pLux to regulate free DnaA(a DNA binding protein that binds to Px- 5 binding sites) concentration in a cell.

pLux transctiptional activity would respond to autoinducer oscillations. pLac promoter is to fine tune the upstream transcriptional activity so that during autoinducer peak, because of pLux transcription, free DnaA is just above the threshold for cell cycle progression.
