Part:BBa_K1031015
NahR-Psal-B0032-RiboJ-CI
Components and Characterization
K1031015 consists of NahR[1], Psal promoter, RBS B0032[2] and transcription factor CƖ. CƖ functions as a repressor of hybrid promoter[3]. It is the repression branch of the Band-pass Filter circuit (Fig 1). In order to fine-tune the expression level of transcriptional repressor, we adjusted the RBS (Ribosome Binding Site) intensity in front of CƖ. K1031015 adopts RBS B0032.
Fig 1 Repression circuit of Band-pass Filter. The orange square represents transcription factor NahR activating Psal promoter shown in light blue. Once Psal promoter is initiated, CƖ repressor, which is shown as square in dark blue, is expressed, inhibit hybrid promoter marked with two colors, thus inhibit expression of sfGFP. The RBS B0032 is shown in light green, regulating the expression level of CƖ protein.
