Design Notes

Primers used in the PCR: 

colicin_FP: gcttctag atgagcggtggcgatg(TM:56C)
colicin_RP: gct ctgcagcggccgctactagta ttacttaccccgatgaatatcaatatgt (TM:56C)

Digested the PCR product with XbaI and PstI, and ligated the digestion product with a gel purified XbaI,PstI digested pSB1A2 plasmid backbone.
