Usage and Biology

  • Include within the ORF of a protein that you need transport into the nucleus (e.g., a synthetic transcription factor)
  • Fluorophore-NLS fusions are handy for making glowing nuclei

PKKKRKV motif from the Simian Virus 40 Large T-antigen. When added to a eukaryotic protein, that protein localizes to the nucleus. in our hands, the localization appears to be very efficient (data coming soon). This tag is very handy for live cell fluorescent labeling, since image processing of glowing nuclei with nice defined edges is much simpler than quantifying diffuse signal in cell bodies with irregular edges.
