pYMB essentials + RBS(Pmsp3) + tetR + RBS(Pmsp3) + GFP

Pmsp1 + RBS(Pmsp3) + tetR + RBS(Pmsp3) + GFP

The Pmsp1 is the second strongest promoter on AMB-1.

RBS(Pmsp3) is the RBS site from Pmsp3, the strongest promoter of AMB-1.(BBa_K624012)

TetR serves as a switch, which can turn on the downstream gene when tetracycline is present; and vice versa.

GFP serves as a reporter to show if the construct work or not.
